The Plasmid DNA Analysis Kit is employed in the automated analysis of supercoiled and linear plasmid DNA. Accurate sizing and relative quantification is provided for supercoiled plasmid between 2,000 and 10,000 bp. High purity supercoiled plasmid is desirable for numerous applications including: QC of purifications, QC of DNA vaccines, and QC for gene therapy products.

Examples

The accurate sizing and relative quantification of supercoiled plasmid DNA depends on the use of a quality DNA ladder. The Plasmid DNA Ladder (FS-SLR940), shown in Figure 1, is comprised of carefully selected and well defined supercoiled plasmids that provide an ideal curve for sizing supercoiled plasmid DNA.

The Plasmid DNA Analysis Kit (DNF-940) excels at separating, sizing, and providing relative quantification of supercoiled plasmid DNA. Analysis of supercoiled Plasmid DNA is essential as a QC step for a variety of applications including: plasmid synthesis, plasmid purification, DNA vaccine manufacturing, and gene therapy development. Supercoiled plasmid DNA is shown in Figure 2.

High resolution separations and relative quantification of linearized plasmid DNA are also produced by the Plasmid DNA Analysis Kit (DNF-940). However, accurate sizing is not calculated, as the Plasmid DNA Ladder (FS-SLR940) is optimized for use with supercoiled plasmid DNA. An example separation of linearized plasmid electrophoresed with the Plasmid DNA Analysis Kit is shown in Figure 3.
