DNA/RNA Oligonucleotide Purity Analysis

Synthetic DNA and RNA oligonucleotides (oligos) are widely utilized as tools in disease diagnostics, medical and pharmaceutical research, and molecular biology. Accurate characterization of the purity of synthesized oligos is imperative, as impurities can significantly affect performance, reproducibility and overall success of DNA- and RNA-based applications.

The Oligo PRO™ system from Advanced Analytical provides a rapid, accurate and economical method for performing high resolution, size-based oligo purity analysis in support of oligonucleotide production, purification and research. Compared to traditional methods, the Oligo PRO™ offers superior separation resolution, higher sample throughput, automation and improved quantification of impurities.

  • Parallel 96-capillary gel electrophoresis with on-line UV absorbance detection (CGE-UV) provides unsurpassed throughput and automation; a lower cost, 24-capillary system is also available for medium throughput applications.
  • CGE-UV is the method of choice for assessing oligo purity, offering superior size-based resolution and nL sampling volumes. Relative purity is determined on-line by directly measuring UV absorbance, and this signal intensity is free from the sequence and size-dependent variations caused by ionization suppression that prove troublesome in mass spectrometry methods.
  • Predefined separation and capillary conditioning methods, and an intuitive user interface help streamline instrument operation.
  • Advanced data analysis software performs manual or automated peak integration and provides customized reports for individual samples or an entire 96-well sample plate.
  • A full line of reagents, specifically optimized for use with the Oligo PRO™ system, provide a cost effective, "turn-key" solution for oligo purity analysis. Single nucleotide resolution can be achieved for up to 100-mer lengths and higher using appropriate sample loading conditions.
  • A unique, four position slide-out stage accommodates up to three 96-well sample plates allowing for unattended analysis of up to 288 samples.
